Documentation: Japanese encephalitis vaccine
Background: Japanese encephalitis virus (JEV) is the leading cause of viral encephalitis in southeast Asia. Although no treatment is currently available, vaccination effectively prevents the disease. In a non-inferiority study, we aimed to compare the safety and immunogenicity of a novel, second-generation, inactivated candidate vaccine for JEV with a licensed, mouse-brain-derived vaccine.

Documentation: Automatic analysis of asymmetric inhibition zones

Background: Reading of agardiffusion tests is currently performed mainly visually, except for inhibition of zones generated by paper discs, test tablets or wells cut into the agar lawn for which several automatic systems are available (e.g. BIOCOUNT). Theses systems are based on image processing principles such as image detection, image smoothening, edge detection, edge enhancing, and area detection. However, all these systems determine and report only the idealized area of the inhibition zone, respectively report the diameter of these almost round (symmetric) inhibition zones.
Aim of this study was to determine if asymmetric (e.g. elliptic or irregularly shaped) inhibition zones can be detected reliably and the included inhibition zone measured automatically. In addition, it was investigated whether from asymmetric inhibition zones of antifungal agents reproducible endpoints (e.g. MIC’s from strip-tests) could be derived.

- Based on the dual telecentric illumination the Bioreader® 5000 does have a better dose/level response (signal/noise ratio) compared to conventional readers.
- Bioreader® 5000 does have a lower background count on non-stimulated cells. Nevertheless it does have a significant higher count on stimulated cells.
- Reference: Internal quality-management study of BIO-SYS performed at an internationaly operating research institute. Up to 4500 spots/well could be counted during the study.
